Nuestro laboratorio tiene como finalidad investigar los mecanismos celulares y moleculares involucrados en la regulación de la respuesta inmune en condiciones fisiológicas y patológicas. En particular investigamos el impacto de la interacción entre proteínas y glicanos en fenómenos de inflamación crónica, autoinmunidad y escape tumoral.

Línea de investigación

En nuestro laboratorio estudiamos la interacción entre proteínas (particulamente galectinas) y carbohidratos; y cómo esta interacción modula la respuesta inmune. Nuestro objetivo es conocer estos mecanismos tanto en el escenario fisiológico y patológico para utilizar esta información con fines terapéuticos. Recientemente determinamos que la galectina-1 (Gal-1), una proteína perteneciente a la familia de galectinas que producen y secretan los tumores, mata los linfocitos T activados cuya función es erradicar al tumor; identificando un nuevo mecanismo de escape tumoral. En este contexto, Gal-1 polariza la respuesta inmunológica hacia un perfil de secreción de citoquinas de tipo Th2. Efectivamente, el bloqueo de la expresión de Gal-1 en el microambiente tumoral genera una respuesta antitumoral de tipo Th1 con altos niveles de IFN-? e IL-2. Demostramos además que la administración de Gal-1 induce una mejoría significativa de las manifestaciones clínicas de enfermedades autoinmunes del tipo Th1 como la artritis reumatoidea, la encefalomielitis autoinmune experimental, la inflamación intestinal crónica y la uveitis autoinmune. La administración de Gal-1 revierte los síntomas de la enfermedad al matar selectivamente a linfocitos Th1. Actualmente investigamos el impacto de la glicosilación diferencial en estos procesos en células T-CD4 + /CD8 + , células dendríticas (CDs), macrófagos y células NK. Estas observaciones posicionan a Gal-1 como un nuevo mediador del escape tumoral, la inmunoedición de tumores y la modulación de la respuesta inmune in vivo lo que nos conducirá probablemente a la identificación de nuevos blancos terapéuticos y de diagnóstico.

Nuestro grupo trabaja en la interface entre la inmunología, la biología tumoral y la glicobiología, utilizando aproximaciones in vitro e in vivo, intentamos entender cómo es que las interacciones moleculares entre las lectinas endógenas y sus ligandos pueden mediar procesos celulares relevantes para la tolerancia inmunológica, la inflamación y la angiogénesis. Por más de 20 años hemos estado investigando el rol de las proteínas de unión a glicanos para mediar procesos celulares centrales para la regulación inmunológica de enfermedades humanas. Nuestro grupo, compuesto por químicos, biólogos celulares y moleculares y bioquímicos, comparte el objetivo común de realizar investigaciones serias y esenciales, así como validaciones pre-clínicas para las estrategias terapéuticas derivadas de nuestros descubrimientos en materia de inflamación, autoinmunidad y cáncer. Nuestro objetivo es contribuir con nuevos descubrimientos que unan la brecha entre la glicobiología fundamental y la inmunología y el desarrollo de nuevos blancos terapéuticos en inflamación, autoinmunidad y cáncer. Nuestros estudios, resumidos en más de 180 publicaciones en revistas internacionales de alto impacto, permitieron la identificación de un nuevo paradigma basado en las interacciones galectinas/glicanos en la homeostasis inmunológica y el diseño de nuevos enfoques en autoinmunidad y cáncer.
